c-Src knock down increases the expression of OXPHOS complexes. A) WT, Src++ (B), and Src (C) cells were transfected with c-Src specific siRNA (c-Src) and control siRNA (cont). The expression of OXPHOS complex subunits were detected by Western blot analyses with respect to GAPDH antibody probing and Ponceau S membrane staining as loading controls. Quantitation of the expression of OXPHOS complex subunits are represented as the mean ± SD of at least three experiments (right panel) and expressed as a percentage of the control siRNA cells (cont = 100%). Significant difference was observed between c-Src and control siRNA transfected cells in the expression of NDUFB8 and COI in WT cells (P = 0.0001 and P = 0.0029, respectively), Src++ cells (P = 0.0020 and P = 0.0056, respectively), and Src cells (P = 0.0004 and P = 0.0356, respectively). Statistical differences were analyzed between the cont and c-Src siRNA transfected cells with the unpaired student’s t-tests (2-tailed), *P < 0.05.
