Figure 6

Whole-mount immunofluorescence imaging of the sciatic nerve for neurofilaments, blood vessels, and myelin proteins.

Representative (5×) and positional Z-stack compact images at 10× from an isografted right sciatic nerve (G-5/7) with saline treatment to show the orientation and distribution of neurofilaments (NF-H, green), myelination (MP0, red), and DAPI (blue), as well as the merged view at different zones of the nerve. White asterisks indicate an abundance of aberrant neurofilaments, and yellow asterisks indicate myelin disruption. Scale bars: 100 µm for 10× images and 100 µm for 5× images. Each image is representative of at least four immunostained whole nerves from four different mice. DAPI: 4’,6-Diamidino-2-phenylindole; G-5/7: 5 mm gap and 7 mm nerve grafting.
