Figure 2.
Global host transcriptomic changes in the presence or absence of a T6SS. A. Heat map analysis summarizing the transcriptomic differences in the presence or absence of a functional Bpm T6SS, showing the 400 most differentially expressed host genes at 12 h post infection. Genes in box 2 indicated an upregulation in the absence of T6SS as compared to WT in box 1. This suggested a transcriptional repression associated to the T6SS (box 2 vs 1). Genes in panel 4 are downregulated as compared to panel 3, suggesting a transcriptional activation associated to the T6SS B. Using ingenuity pathway analysis, hallmark gene association suggests a role for the most differentially expressed genes to be associated with TNF-α signaling via NF-κB. In addition, gene ontology analysis suggests a role of differentially expressed genes linked to cell proliferation, cell death, among other metabolic processes. C. The volcano plot demonstrates specific differential expression of genes during Bpm IEC infection generated with p-values less than 0.05 in the presence (Bpm WT) or absence of a functional T6SS (Δhcp1).
