Cell distribution of WT and N-glycosylation mutants of CD98. Cells were transiently transfected with WT or mutant constructs. After 24 h, cells were fixed onto the slide and stained with anti-CD9818 that recognizes the epitope protruding towards the extracellular side. After extensive washing the samples were permeabilized using Triton X100 and stained with the anti-FLAG that recognizes the tag located at the N-terminus33. Samples were then analyzed by confocal microscopy. Confocal images showed a single, representative, section of a Z-series taken through the entire cell. WT and single mutants were shown in panel (A), while double (Dm: N381/424Q), triple (Tm: N365/381/424Q), and quadruple (Qm: N365/381/424/506Q) mutants were shown in panel (B). Scale bar 10 µm.
