Esomeprazole reduced expression of markers of endothelial dysfunction in response to tumour necrosis factor (TNF)-α, compared with vehicle control in primary HUVECs. Expression of two markers of endothelial dysfunction, endothelin-1 (ET-1) (A), and vascular cell adhesion molecule-1 (VCAM1) (B) was increased in HUVECs treated with TNF-α (10 ng/mL) after 24 h (control + TNF-α, green); HUVECs not treated with TNF-α (control, grey) have significantly lower expression of markers assessed. Both esomeprazole magnesium trihydrate (Eso MTH) and esomeprazole magnesium hydrate (Eso MH) at 100 μmol/L abolished the increase in both ET-1 (A), and VCAM1 (B) mRNA expression in response to TNF-α.
