Peptide major histocompatibility complex (pMHC) tetramer staining of HLA-A*01:01–restricted Epstein-Barr virus (EBV)–specific T cells. Peripheral blood mononuclear cells (PBMCs) from 6 healthy EBV+ HLA-A*01:01–positive donors were incubated with different pMHC tetramers that were predicted to be strong binders specific for EBV and restricted to HLA-A*01:01. A, Representative examples for donor B are shown. Total PBMCs were stained with pMHC tetramers and CD8. Tetramer-positive cells were sorted and expanded for 2 weeks. Only Epstein-Barr virus Latent Membrane Protein 2 specific T cells targetting the ESEERPPTPY peptide (EBV-LMP2ESE) could be expanded.
