Effects of AYC-EV administration on T cell responses in an OVA-induced asthma model. (a) Splenic T cells isolated from mice (at 2 days after the last OVA challenge) of PBS-, OVA-, OVA + Dex-, OVA + AYC-EV (8 mg/kg)-administrated groups (n = 4 mice per group) were stimulated in vitro with Cell Stimulation Cocktail (PMA/Ino) for 4 h. Following incubation for 4 h, T cells were immunostained for dead cells (L/D), CD3, CD4, IFN-γ, IL-5, and IL-17A as described in the material and methods section, and percentages and absolute numbers of Th1 (CD3+CD4+IFN-γ+ cells), Th2 (CD3+CD4+IL-5+), Th17 (CD3+CD4+IL-17A+ cells), and mixed Th2/Th17 cells (CD3+CD4+IL-5+IL-17A+ cells) were analyzed using flow cytometry. (b) Splenic T cells isolated from mice from each group were immunostained for L/D, CD3, CD4, CD25, and Foxp3 as described in the material and methods section, and percentages and absolute numbers of regulatory T cells (CD3+CD4+CD25+Foxp3+ cells) were analyzed using flow cytometry.
