In the initial step (1) the backbone phosphate of Am may be bound by H321 in interface loop; then, Y247 in the Gate Loop1 interacts with N6 of Am (2), followed by hydrogen binding from D233 and P234 and methyl group transferred from SAM to N6 of Am (3). Finally, N6-methylated Am is released from the pocket due to the steric clash (4).
