Methylation by 2′-O-ribose of different sequences prevents TLR7 activation. FLT3L DCs from wt or Tlr7^−/− mice were stimulated with 1 µM CpG ODN 2216, 5 µg/mL RNA40, and RNA66 derivatives or RNA28 derivatives at a final concentration of 10 µg/mL. Supernatants were harvested 20 h p. s. and concentrations of (a) IFN-α and (b) IL-6 were determined in ELISA.
