Tissue clearing overview

(A–C) Images showing the stages of tissue clearing, scale bars: 3 mm. The initially opaque spinal cord (A) becomes transparent after delipidation with CUBIC-L (B). The sample will reach maximum transparency after RI-matching in CUBIC-R+(M) (C).

(D–G) (D) A 1 mL syringe is used as a mold for the sample by first (E) removing the tip. 2% CUBIC-R+(M) agarose is pipetted into the syringe (F) and a cleared sample is embedded in the agarose (G).

(H) After gel polymerization, the embedded sample can be lowered for imaging.

(I) A syringe prepared for gel embedding.
