Phosphorylation of UL97 serine 13 increases its interaction with 14-3-3 proteins. U-2 OS cells were transfected with an expression vector for either an HA-tagged S13A mutant or a kinase-dead derivative of UL97 (0.7 μg) together with expression vectors for the indicated Flag-tagged 14-3-3 isoform (0.6 μg) and expression vectors for either a V5-tagged S13A mutant or kinase-dead derivative of UL97 (0.7 μg). After 48 h, lysates (Input) were harvested, subjected to immunoprecipitation with anti-HA antibody (IP: anti-HA), and analyzed by Western blot with the indicated antibodies.
