Loss of MirB disrupts the uptake of [Fe]DAFC-FITC in A. fumigatus. Fungal strains were grown in iron-depleted minimal medium for 16 h and subsequently incubated with 5 μM [Fe]DAFC-FITC for 30 min before imaging by epifluorescence microscopy. Bar, 10 μm. Pictures were taken with the same settings (not auto), indicating that the increased ambient fluorescence of the ΔsidA ΔmirB strain compared to the ΔsidA and ΔsidA ΔmirB strains reflects the lack of [Fe]DAFC-FITC uptake.
