Effects of charge conserving mutation of R2 on the closed state of the channel

(A and B) Representative tail currents recorded from WT (A) or R2K (B). The tail currents were elicited by a prepulse to 100 mV, in 10 mV decrements from 0 to −60 mV.

(C) The R2K mutation negatively shifted the G-V relationship of the channel. Lines indicate fits of the data to a Boltzmann function.

(D) The deactivation (channel closing) time constant Ʈdeact in WT or R2K mutation. Ʈdeact was obtained from exponential fit to the tail currents. Proton currents were recorded in HEK293 cells expressing Hv1 mutations, pHi = pHo = 6.0. Data are represented as mean ± SEM.
