The effect of curcumin on the elevated mRNA transcript levels of copper transporters CTR1 and ATP7A in MCF-10A-Cu cells relative to their parental MCF-10A cells. Total RNA was extracted utilizing TRIzol reagent (Invitrogen, Carlsbad, CA, USA) as directed by the manufacturer. As described in Section 2, CTR1 and ATP7A mRNA expression was quantified using real-time PCR. To determine the effect of curcumin on mRNA expression, only MCF-10A+Cu cells (normal MCF-10A cells grown in a medium containing 25 µM CuCl2) with enhanced mRNA expression of copper transporters were treated with 25 µM curcumin. Values reported are ±S.E.M. of three independent experiments. * p value < 0.05 when compared to control.
