miR-375 inhibits ESCC cell growth by directly targeting PRDX1. (A) The relative mRNA levels of PRDX1 in TE-1/ECA-109 cell co-transfects of a miR-375 mimics and pcDNA3.1-PRDX1 vector for 48 h. (B) Using a clonogenic assay, we determined the effect of PRDX1 overexpression and miR-375 on TE-1/ECA-109 cell colony development. (C,D) A CCK-8 assay was used to evaluate cell proliferation in TE-1/ECA-109 cells that had been transfected with miR-375 or a sham control, with or without PRDX1 overexpression. (E,F) A transwell assay was used to analyze cell invasion following transfection with miR-375 or a sham control in TE-1 and ECA-109 cells, with or without PRDX1 overexpression (0.2% crystal violet staining, original magnification 400×). (G,H) To detect apoptosis, a flow cytometry assessment was conducted using a mock control or miR-375 overexpression TE-1/ECA-109 cells, with/without PRDX1 over-expression.
