The conserved jaw joint regulatory sequence 1 (JRS1) enhancer was first identified as a conserved non-coding element (CNE) in silico, then confirmed with transgenesis experiments of a range of species-specific JRS1 sequences driving fluorescent reporter expression in the jaw joint. Finally, the JRS1 enhancer was deleted from the zebrafish genome to reveal a transient jaw joint dysmorphology and partial fusion. m: Meckel’s cartilage; pq: palatoquadrate; ra: retroarticular process.
