L. donovani attenuates serine phosphorylation of Elk-1. Cells were incubated with or without amastigotes of L. donovani for 17 h and then stimulated with 100 nM PMA for 15 min. Detergent lysates were prepared with modified RIPA buffer as described in Materials and Methods, and soluble proteins were separated by SDS-polyacrylamide gel electrophoresis (7.5% polyacrylamide) followed by transfer to nitrocellulose membranes. The membranes were probed with phospho-Ser³⁸³-specific Elk-1 antibody. The blots were developed by ECL, and an autoluminogram is shown. The data shown are representative of two independent experiments that gave similar results; values in the histogram represent the average.
