Conjugation of P3on the surface of E91NP. E91NPs were used to conjugate P3 on the surface using EDC–NHS chemistry. The detailed protocol for conjugation has been described in the Experimental procedures section. The conjugation was confirmed by several techniques. A and B, DLS plots show the changes in (A) ζ-potential (data compared with ζ-potential of blank NPs from Fig. 1A); (B) DLS size before and after P3 conjugation (data compared with a hydrodynamic diameter of blank NPs from Fig. 1B); (C) FTIR spectra showing the presence of C=O stretch of saturated ester (1742 cm−1) and 2° amide (1680 cm−1), imparted by the conjugation of P3 molecules on the surface of NPs. Biotinylated P3 (bio-P3) was used for isothermal titration calorimetry (ITC). D, heat flow profile of ITC exhibiting time upon injection of HRP–streptavidin to NPs surface-functionalized with either (i) P3 or bio-P3. DLS, dynamic light scattering; EDC, N-(3-dimethylaminopropyl)-N′-ethylcarbodiimide hydrochloride; HRP, horseradish peroxidase; NHS, N-hydroxylsuccinimide; NP, nanoparticle; ζ-potential, zeta potential.
