Figure 9

Overnight chow intake in male Het Gcg-Cre rats (N = 14) with bilateral cNTS-targeted injections of Gq DREADD virus after i.p. injections of saline followed by saline (control, saline–saline), saline followed by CCK (saline-CCK), CNO followed by saline (CCK-saline), or CNO followed by CCK (CNO-CCK). A, average food intake over the 12 h dark cycle grouped by i.p. treatment combination. Compared to overnight food intake after saline–saline treatment, CNO-saline appeared to inhibit food intake after dark hour (D)4, although the cumulative hypophagic effect of CNO-saline did not reach significance until D12 (∗P = 0.045 vs. saline–saline). Saline-CCK did not suppress food intake at any timepoint compared to intake after saline–saline. However, a significant hypophagic effect of CNO-CCK was evident at D4 (P = 0.022), D6 (P = 0.0008), and D12 (P = 0.017). B, individual within-subjects data for cumulative intake by rats after each i.p. injection condition at dark hour (D)4, D6, and D12, with intake expressed as % reduction compared to intake by the same rat after control (saline–saline) treatment. CNO-CCK suppressed food intake to a greater degree than CCK alone at D4 (∗P = 0.013) and D6 (∗P = 0.011), and suppressed intake more than CNO alone at D6 (∗P = 0.012).
