Combination treatment challenge against A. xylosoxidans invasion in BEAS-2B + 20% ASMDM-1 cell model. Confluent BEAS-2B cells were washed and infected with OD600 0.1 ± 0.02 for one hour to allow for intracellular invasion. Following this, cells were washed thrice with 100 μg/mL gentamicin before addition of combinations in a final concentration of 20% ASMDM-1 and 80% DMEM medium for three hours. Cells were washed twice with 1 × PBS before being lysed and intracellular bacterial loads were enumerated using CFU/mL. (a) ATCC 27061 and (b) AX 4365. Correlation with planktonic synergy was determined previously and is denoted as (S) synergy and (I) indifferent. (*) indicates the results are statistically significant (p ≤ 0.05) (*) based on unpaired Student’s t-tests (with Welch’s correction). Experiments were performed in biological triplicate (n = 3).
