Figure 1: Analysis of immune cell subpopulations in early stage of DKD in OVE26 mice.

Figure 1:

(A) Schematic diagram illustrating the experimental workflow. Phosphate-buffered saline-perfused kidneys of control wildtype (WT) and diabetic OVE26 mice of 3 months of age were isolated and dissociated. CD45+ cells were sorted from each sample by flow cytometry were pooled from 3 mice per experimental group for scRNAseq analysis. (B) Uniform manifold approximation and projection (UMAP) of all CD45+ cells from WT and OVE26 mouse kidneys. (C) Proportions of CD45+ immune cell subtypes in WT and OVE26 kidneys are shown as a bar graph. (D) UMAP plot of MNPs from WT and OVE26 mouse kidneys. (E) Proportions of each MNP subclusters in WT and OVE26 kidneys. DC, dentritic cell; Mac, macrophage; cDC, conventional DC; pDC, plasmacytoid DC; ILCs, innate lymphoid cells; NK&T, natural killer and T cells; IFNhi, interferon (IFN)-induced gene expression-high; Mrc1hi, Mannose receptor C-type 1 expression-high; Trem2hi, triggered receptor expressed on myeloid cells 2 expression-high; Ccr7hi, C-C motif chemokine receptor 7 expression-high.
