Northern blot analysis of WdCHS4 expression. Total RNA (20 μg) was prepared from wild-type W. dermatitidis 8656 (lanes 1 and 4) and the temperature-sensitive morphological mutants Mc3 (lanes 2 and 5) and Hf1 (lanes 3 and 6) grown at 25 and 37°C, respectively, for 24 h and electrophoresed in a formaldehyde-containing 1.2% agarose gel before transfer to a nylon membrane. The membrane was probed with a ³²P-labeled 0.8-kb BamHI fragment of WdCHS4 and a 0.6-kb PCR fragment of the actin gene (WdACT1) of W. dermatitidis, as indicated.
