Mass spectral analysis of native and thio-linked hemi-A heparosan

Product GlcNAc-R’ (GlcNAc-GlcA)1-GlcNAc-R’ (GlcNAc-GlcA)2-GlcNAc-R’
Native (= 4-OH)
Observed 950.38 ± 0.11 1,329.50 ± 0.04 1,708.65 ± 0.05
Predicted 950.29 1,329.40 1,708.51
4-SH Analog
Observed 966.29 ± 0.11 1,361.43 ± 0.05 1,756.56 ± 0.06
Predicted 966.27 1,361.36 1,756.44
Observed ∆ S-O 15.91 31.93 47.91

The observed and predicted masses (Da) are shown. The mass difference between the natural O-linked and the artificial S-linked species (last row of table) corresponds to the additional mass (~16 Da per S atom) associated with the replacement of oxygen with 1, 2, or 3 sulfur atoms (acceptor R’ = GlcA-GlcNAc-GlcA-C2H4-amido-benzaldehyde).
