Lane 1 shows the molecular weight (MW) markers (in thousands), and lane 2 shows the background detection of a total trypanosome extract by the serum from non-infected mice. Lanes 3, 4, and 5 respectively contain total trypanosome extract, purified VSG, and purified trypanosome HSP60, stained with serum isolated from infected mice. (b) Quantification of the trypanosome HSP60 by the dot blot technique. A serial dilution of purified HSP60 (50 μl) was used to obtain a standard for densitometric analysis; 25 μg of total trypanosome extract was spotted per dot.
