(A) BMMCs were incubated with either 2 μM PD184352 or 1 μM VX745 as indicated for 1 h before stimulation with 10 ng/ml IL-33 for a further 1 h. Total RNA was isolated and the induction of granzyme C mRNA determined by qPCR. For comparisons with the IL-33 only condition, a P<0.05 is indicated by *, and P<0.01 by ** (Brown-Forsythe and Welch ANOVA followed Dunnett’s T3 post hoc testing). (B,C) BMMCs were incubated with either 2 μM PD184352 or 1 μM VX745 as indicated for 1 h before stimulation with 10 ng/ml IL-33 for 24 h. Cells were then fixed, stained for granzyme C, and analysed by flow cytometry. Representative histograms are shown in (B) and quantification of the geometric mean in (C). Graphs show the mean and standard deviation of three independent stimulations. For comparisons to the IL-33 only condition, a P<0.05 is indicated by *, P<0.01 by ** and P<0.001 by *** (one-way ANOVA followed Dunnett’s T3 post hoc testing).
