(A) Independent cultures of BMMCs from four wild-type and four MSK1/2 mice were isolated and stimulated with 10 ng/ml IL-33 for 0, 1, or 2 h. Total RNA was isolated and the induction of granzyme C mRNA determined. Fold-change was calculated relative to the level in unstimulated wild-type BMMCs. An average of GAPDH and 18s levels were used to correct for loading. (B,C) Wild-type and MSK1/2 knockout BMDMs were stimulated with 10 ng/ml IL-33 for 24 or 48 h and the levels of intracellular granzyme C determined by flow cytometry. Representative plots are shown in (B) and quantification of the geometric mean from three mice per genotype in (C). For (A) and (C), graphs show the mean and standard deviation with individual cultures shown as symbols. For comparisons with the IL-33 only condition, a P<0.001 is indicated by *** (two-way ANOVA followed Sidak’s post hoc testing).
