PCK1 is expressed in human ß-cells exposed to high glucose and T3 concentrations. Human islets were transduced with adenovirus expressing ZsGreen under the rat insulin promoter. Islets were dispersed and cultured in low (6 mM) or high (20 mM) glucose concentrations. After 48 h, cells were collected and sorted by FACS to separate β-cells from non-β-cells. mRNA was extracted and qPCR was performed to assess the levels of insulin, PCK1, ChREBPα or ChREBPβ.
