Elaiophylin induces intracellular and mitochondrial ROS in A549 cells. A549 cells were exposed to elaiophylin for 3 h with or without pre-treatment with NAC (10 μM) for 6 h. (A,B,D,E) Intracellular ROS level detection was assessed using DCFH-DA fluorescence probe; while mitochondrial ROS level detection was assessed using Mitosox red mitochondrial superoxide probe. (C,F) Cytochrome C translocation was determined by Western blot analysis. GAPDH and VADC1 were used as reference proteins in cytoplasm and mitochondria, respectively. Data were expressed as means ± SD of three experiments and each experiment included triplicated repeats. ** p < 0.01 vs. control, ## p < 0.01 vs. Elaiophylin group.
