Different strategies of protein suppression. (a) A small-molecule inhibitor binds to the active site of a protein of interest (POI) to inhibit its enzymatic functions. Most enzyme inhibitors bind noncovalently and reversibly. (b) PROTAC directs E3 ligase to a POI and mediates the polyubiquitination of the POI by an E2 conjugating enzyme. The modified POI is recognized and degraded by the 26S proteasome. The PROTAC can then be recycled to induce the next round of the POI degradation.
