The modality simplifies the optimization of PROTACs. (b) The dTAG molecule is composed of an E3 ligase ligand linked to a POI ligand, which facilitates formation a ternary complex between the fusion protein and E3 ligase, leading to polyubiquitination and degradation of the POI. The key difference between the dTAG and aTAG technologies is the identity of the TAG protein used. (c) Misfolded proteins are degraded through recognition of hydrophobic patches.
