When neutrophils migrate from systemic circulation to the pulmonary vasculature in ALI mice, the effect of mechanical deformation and the characteristics of the microenvironment make them more adaptive to defense against pathogens (Prok2hi Neu). During subsequent trans-endothelial and -epithelial movement, large numbers of neutrophils develop into the Fth1hi subset, with increased resistance, delayed apoptosis and greater production of inflammatory mediators. Fth1hi Neu exhibited lower expression of L-selectin (CD62L) and C-X-C motif receptor 2 (Cxcr2), and higher expression of Cxcr4 and intercellular adhesion molecule-1 (Icam1).
