Interaction of single molecules of chimeric sfGFP-VASH2–SVBP complex bearing the N-terminal region of VASH1 [V2_FL(NtV1)] with Taxol-stabilized microtubules enriched in tyrosinated HeLa tubulin studied by single-molecule TIRF microscopy. The presence of the acidic N-terminal region of VASH1 strongly changed the behavior of VASH2–SVBP. The chimeric complex (50 pM, green) showed shorter residence time and recovery of diffusion capacity on microtubules (magenta), resembling full-length VASH1 complex. Scale bar, 2 μm. Images were collected every 50 ms for 45 s and films are 2.5× accelerated.
