Groups of BALB/c mice (n = 4–8) were immunized i.m. twice with 1 × 108 PFU MVA-S, MVA-ST, or PBS as negative controls. (A–D) Splenocytes were collected and prepared on day 14 after boost immunization and stimulated with the H2-Kd–restricted peptide S268–276 (S1; GYLQPRTFL) and tested using ELISPOT assays and ICS FACS analyses. (A) IFN-γ+ SFC measured by ELISPOT assays. (B and C) IFN-γ–producing CD8+ T cells measured by FACS analysis.
