Quantification of ssDNAs in lysates from vector-transfected cells. Mouse FL-10 cells were transfected with either the pssXA plus pssXB(AG30) vector set or with the synthetic ODN, AG34 (both via lipofection), and 24 h later the cells were lysed for isolation of low molecular weight DNA. The samples were analyzed for the presence of the predicted ssDNA product or the presence of ODN by primer extension with a 15 nt primer 5′ end-labeled with ³²P. Parallel samples were spiked with AG34 at concentrations calculated to mimic 10⁴–10⁷ molecules per cell. Lanes 1 and 2 contain 5′ end-labeled synthetic oligonucleotides of length 30 and 41 as size markers.
