Cmpd-3 can potentiate the NE-mediated cAMP response in transfected Rat-1 fibroblasts (A), human SK-N-MC, a transformed neuroblastoma cell line (B) or primary mouse cardiomyocytes in a PAM assay (C). To show PAM effects of potency and Emax, the concentration of NE is held constant at its Emax (10−4 ​M) (red line). Cmpd-3 is then added in a dose-response (blue line). Any signal above the red line is potentiation. Cmpd-3 potentiates the NE-mediated cAMP response with a potency of 0.1 pM in neuroblastoma cells (A) or 0.5 ​nM in primary cardiomyocytes (B).
