Under the signal of necroptosis, Brd4 is recruited to the promoter of MLKL and forms a transcription complex with IRF1, P-TEFb, and RNA polymerase II to regulate the expression of MLKL, which leads to necroptosis. BET inhibitors such as JQ1 interferes with the transcription complex formation.
