(A) Representative G4 motif and formation, G-tracts in bold and loops denoted. (B-E) The proportion of errors (detectable & undetectable) within each G4 motif was analyzed to determine the polymerase error distribution between G-tracts and loops. G-tracts = error in G-tract only (may include insertions >1nt), Loops = error in loop only, G-tracts & Loops = errors that occur in both a G-tract and adjacent loop (e.g., deletion of >1 nt). Errors that could not be unambiguously assigned to the G4 were not counted in this analysis. Tandem mutations were counted as independent errors. Fisher’s Exact Test for a 2x3 or 3x3 contingency performed on error counts, p-value indicated where statistically significant. N = total error counts from two to three independent polymerase reactions (See Table S2).
