Depletion of neutrophils did not impact bacterial load in central or peripheral tissues of Ot-infected mice. B6 mice (n=10/group) were i.p. injected with anti-Ly6G, anti-Gr-1 or anti-Ly6G/6C antibody (clone#1A8, RB6-8C5 and NIMP-R14, respectively; 250 µg/mouse) at day -1, 0, 1 and 2 post-infection. Control mice were treated with IgG (250 µg/mouse). At day 0, mice were i.d. infected with Ot (4×103 FFU) in the ears. (A) On day 3, skin tissue and draining lymph nodes (dLN) were collected for quantification of bacterial load. (B) Bodyweight following Ot infection. (C) Lung and brain tissues were collected at days 7 and 14 and bacterial load was quantified. Values are shown as mean ± SEM from single experiments and are representative of two independent experiments. A one-way ANOVA with a Tukey’s multiple comparisons test is used for statistical analysis. A two-tailed Student’s t-test was used for comparison of two groups. No significant difference was found in these results.
