(a) p62/SQSTM1, herein shortened as p62, triggers a basal Keap1 proteasomal degradation (yellow dots) controlling NRF2 stabilization/activation. (b) Autophagy activation degrades p62 (left part, blue dots), stabilizing Keap1 that can bind NRF2 and induce NRF2 degradation (right part, purple dots). (c) When autophagy is compromised (red cross), p62 accumulates and can bind Keap1 and trigger Keap1 proteasomal degradation (yellow dots), inducing the stabilization of NRF2.
