In vitro analyses of gastric cancer (GC) cell lines subjected to Methyltransferase-like 3 (METTL3) siRNA transfection. (A) Semi-quantitative RT-qPCR analysis to detect METTL3 gene expression in GC cell lines. (B) METTL3 expression was significantly suppressed by METTL3 siRNA transfection in both MKN7 and NUGC3 cell lines. GC cells were transfected with either METTL3-siRNA (siMETTL3) or negative control siRNA (Negative CTRL), and expression ratio of siMETTL3 to Negative CTRL cells was measured at 24 and 48 h after transfection by RT-qPCR. (C and D) Proliferative effects of METTL3 in GC cells at 48 and 72 h. The proliferation potential was significantly suppressed after METTL3 knockdown compared with Negative CTRL MKN7 (C) and NUGC3 (D) cells.
