(A) The topology of human SMO protein. (B–E) Cholesterylation of the SMO variants. HEK293T cells were transfected with the indicated SMO-expressing plasmids. After incubation with the medium containing 2 μg/mL cholesterol probe (CP) for 16 h, the cells were harvested and subjected to the click assay with biotin-alkyne. The CP-modified SMO was pulled down with neutravidin beads and analyzed by immunoblotting with the anti-SMO antibody.
