Characterization of the pgm insertion mutation. (A) The mini-Tn5 insertion (5.0 kb) was found to have occurred 604 bp downstream of the pgm methionine start codon. (B) Southern blot analysis of pgm mutants and other Bordetella spp. utilizing the 300-bp PCR fragment (PCR-1), amplified adjacent to the mini-transposon insertion (see panel A). The pgm gene exists in an EcoRI fragment of 8.4 kb. An EcoRI site located on the transposon results in a restriction fragment 539 bp shorter than that of the wild type.
