Febuxostat facilitates nuclear translocation of Nrf2 and degradation of Keap1 in MC3T3-E1 pre-osteoblastic cells. (A) MC3T3-E1 cells were cultured with Febu for the indicated time period. Nrf2 levels in the nucleus were analyzed by western blotting. p84 served as a loading control. Relative changes in the band intensities normalized by respective loading controls are indicated. (B) mRNA expression of Ho-1 and Nqo1, the target genes of Nrf2, was determined using real-time PCR. Gapdh served as an endogenous control to normalize each sample.
