Flow cytometry of EpMab-37-mG2a-f against the human colorectal cancer cell line, Caco-2, and Caco-2 cells in which EpCAM was knocked out (BINDS-16). (A) Caco-2 and BINDS-16 cells were treated with EpMab-37-mG2a-f (red) or buffer control (black), followed by Alexa Fluor 488-conjugated anti-mouse IgG. Fluorescence data were analyzed using the SA3800 Cell Analyzer. (B) Determination of the binding affinity of EpMab-37-mG2a-f for Caco-2 cells using flow cytometry. Caco-2 cells were suspended in 100 µl serially diluted EpMab-37-mG2a-f. Alexa Fluor 488-conjugated anti-mouse IgG was then added. Fluorescence data were collected using the BD FACSLyric and the KD was calculated by GraphPad PRISM 8. EpCAM, epithelial cell adhesion molecule; KD, dissociation constant.
