Upregulation of GRP78 attenuates sensitivity to cisplatin in HNSCC cell line. (A) SCC15 cells were treated with increasing doses of AA147, or dimethyl sulfoxide (DMSO) as control, for 48 hr and the GRP78 protein level was determined by Western blot, with GAPDH serving as loading control. (B) SCC15 cells were treated with either 10 μM AA147 or 300nM of the ER stress-inducer thapsigargin (Tg) for 48 hr. The GRP78 and ATF4 protein levels were determined by Western blot, with GAPDH serving as loading control. (C) Representative images of SCC15 cells treated with 12 μM cisplatin or 10 μM AA147 alone or in combination for 48 hr. Scale bar = 50 μm.
