Lumateperone altered hippocampal microglia under inflammatory conditions. Lumateperone normalized inflammatory markers in microglia isolated from rat hippocampus 16 h after treatment with LPS. A, Adult rats received a subcutaneous injection of either LPS (500 µg/kg diluted in 0.9% saline) or vehicle. Sixteen hours later, rats were injected intraperitoneally with either lumateperone (3 mg/kg) or its vehicle (5% DMSO, 5% Tween 20, 15% PEG 400, and 75% water). B, Hippocampi were dissected and enriched for microglia via Percoll gradient. C, Results show hippocampal microglia levels of Il1b, Tnfa, Il6, Nlrp3, and Csf1r mRNA normalized to control group using the Dct method.
