Zebrafish were treated with 7 μM DMF during the pharyngula, hatching, and protruding-mouth stage for 6 hours and then immediately fixed and Nrf2a protein was labeled via Immunohistochemistry (IHC). The colocalization analysis was performed on 40x confocal images of the liver, and a representative image of the pancreatic islet was taken from the Z-stack. The Pearson’s R coefficients of control (DMSO WT) embryos were converted to a normally distributed A) Z-scores and are shown as means ± SEM. Representative images, zoomed in to show individual cells of the pancreatic islet (yellow circle) and liver, are also shown to demonstrate Nrf2a protein (red) localization near the nuclei (blue). Calculations were performed using a one-way ANOVA followed by Fisher’s LSD post-hoc test. N = 6–12 fish. n.p. indicates timepoints when liver was not present.
