Crosstalk between HH signaling and the glucocorticoid receptor NR3C1 in T-ALL cells: (a) in the absence of glucocorticoids, GLI1 is associated with HDAC1 and transcription of GLI1-regulated genes is active; (b) upon treatment with glucocorticoids, NR3C1 translocates to the nucleus, thereby displacing HDAC1 and facilitating association of the histone acetylase PCAF with GLI1. PCAF acetylates GLI1, which is then targeted for proteasomal degradation.
