Pharmacological inhibition of CHK2 blocks baicalein-mediated G2/M cell cycle arrest. (A) SKOV-3 cells were pretreated with 30 μM CHK2 inhibitor (Chk2 inhibitor II) for 2 h and then with 100 μM baicalein for 24 h. The protein levels of phosphorylated CHK2 (Thr68) and total CHK2 in cells after treatment were measured by immunoblot analysis. (B) SKOV-3 cells were pretreated with 30 μM CHK2 inhibitor (Chk2 inhibitor II) for 2 h and then treated with 200 μM baicalein for 24 h. The percentage of cells accumulated in the G2/M phase after treatment was assessed by flow cytometry. Significance was defined as *, p < 0.05 and **, p < 0.01.
