Spores of the ΔacpA or acpB mutants (top panel) were seeded into 100μl of sDMEM as is or supplemented with0.75% HCO₃^- or 10% NRS and incubated at 37°C in an ambient or 10% CO₂ atmosphere (as indicated on the right) for 24h. Capsule was imaged by India ink negative staining (capsule presence forms a typical bright outer layer).
